چکیده

In a continuing effort to develop inexpensive source tracking methods to detect human fecal contamination in environmental waters, we combined targeted sampling with fluorometry. Targeted sampling works by identifying hotspots of fecal contamination through multiple samplings over everdecreasing distances. Fluorometry identifies human fecal contamination by detecting optical brighteners, primarily from laundry detergents. On St. Simons Island, targeted sampling and fluorometry identified two hotspots of fecal contamination. One hotspot was confirmed as fecal contamination from humans, but the other was not, most likely because of background organic matter fluorescence. Adding a 436-nm emission filter to the fluorometer reduced this background fluorescence by >50%, and with this filter in place, the second hotspot was identified as fecal contamination from birds. As long as a fluorometer is equipped with a 436-nm emission filter, targeted sampling combined with fluorometry may be a relatively inexpensive method to identify human fecal contamination in water.

Combining targeted sampling and fluorometry to identify human fecal contamination in a freshwater creek.

Many bacterial source tracking (BST) methods are too expensive for most communities to afford. In an effort to develop an inexpensive method of detecting human sources of fecal contamination in a freshwater creek during baseflow and stormflow conditions, we combined targeted sampling with fluorometry. Targeted sampling is a prelude to BST and finds sources of fecal contamination by continued sa...

Origin of fecal contamination in waters from contrasted areas: stanols as Microbial Source Tracking markers.

Improving the microbiological quality of coastal and river waters relies on the development of reliable markers that are capable of determining sources of fecal pollution. Recently, a principal component analysis (PCA) method based on six stanol compounds (i.e. 5β-cholestan-3β-ol (coprostanol), 5β-cholestan-3α-ol (epicoprostanol), 24-methyl-5α-cholestan-3β-ol (campestanol), 24-ethyl-5α-cholesta...

Identifying sources of fecal contamination inexpensively with targeted sampling and bacterial source tracking.

Most bacterial source tracking (BST) methods are too expensive for most communities to afford. We developed targeted sampling as a prelude to BST to reduce these costs. We combined targeted sampling with three inexpensive BST methods, Enterococcus speciation, detection of the esp gene, and fluorometry, to confirm the sources of fecal contamination to beaches on Georgia's Jekyll and Sea Islands ...

Microbial Source Tracking

Microbial contamination affects a large number of water bodies in the US. Because it is not feasible to directly monitor all pathogens, it is standard practice to monitor fecal indicator bacteria (FIB), such as total and fecal coliforms, Clostridium perfringens, Escherichia coli, and fecal enterococci, in water. Epidemiological studies have established human health standards based on exposure t...

Field-testing Enterococcus Faecalis for Bacterial Source Tracking

A waste stabilization pond was suspected of causing fecal contamination to Holt Branch, a tributary of Juniper Creek, near Byron, Georgia. We used ribotyping, a genotypic (DNA-based) bacterial source tracking (BST) method, to match Enterococcus faecalis isolates from Holt Branch and the waste stabilization pond. Fecal enterococci in the effluent from the waste stabilization pond averaged 13,863...
